Normalization with Spike
A spike can be added to samples to control sample specific inhibition of the reverse transcription and the qPCR reaction. The spike is added to all samples and to an uncomplicated reference sample. The RT/qPCR efficiency or recover rate of the spike in the reference sample is either known, or assumed to be 1 (=100%). GenEx corrects the Cq values of the other samples based on the difference between the spike's Cq values in the samples and the reference sample.
To normalize the expression of the target genes to a spike, select Normalization with Spike in the Pre-processing menu in the Data editor. This opens a dialog box where you select the row that contains the reference sample and the column (gene) that contains the spike. Specify the RT/qPCR efficiency and if you want the reference sample to be removed. The data is normalized when Apply is pressed, and the spike column removed as well as the reference sample if the check box was ticked.
